Oxidative and pro-inflammatory lung injury induced by desflurane inhalation in rats and the protective effect of rutin

Background Desflurane is a mainstay of general inhaled anesthetics with methyl ethyl ether structure and widely used in clinical practice. It has been reported to induce inflammation lipid peroxidation rat pulmonary parenchyma, increase alveolar macrophages, cause peribronchial infiltration edema. Rutin, flavonoid vitamin P1, known have biological properties including acting as an antioxidant, anti-inflammatory, inhibitor bronchoalveolar polymorphonuclear leukocyte (PNL) infiltration. Objectives The aim this study examine the effects rutin on desflurane-induced injury using biochemical histopathological methods. Material methods rats were divided into 3 groups (n = 6 each): healthy control (HC), rutin+desflurane-treated (DRT) desflurane-only (DSF). Briefly, 50 mg/kg was given orally DRT group equal volume normal saline DSF HC groups. After 1 h, anesthesia induced maintained for 2 h. had sacrificed, lungs removed. Malondialdehyde (MDA), total glutathione (GSH), tumor necrosis factor alpha (TNF-?), nuclear kappa B (NF-?B) levels measured excised lung tissue. removed tissues also fixed 10% formalin, obtained sections stained hematoxylin eosin (H&E) evaluated under light microscopy. results compared those from Results increased MDA, TNF-? NF-?B, decreased GSH PNL infiltration, hemorrhage, damage, edema observed tissue group. Rutin histopathologically shown protect oxidative stress by preventing oxidant parameters decrease antioxidants. Conclusions suggest that may be useful treatment desflurane-associated injury.
